Is it possible to interrupt playing an audio by speech?

if you mean that while the IVR is playing an audio file to the caller, that if the caller says something the audio stops playing?
This can only be done if you are using ASR. As you state you are using DTMF only, then the answer to your question is no.
It cannot be done.
